I know on a full size or compact Glock a Gen 2.5 model has the thumb groove but no rail. Since the subcompact G26 and G27 have no rail anyway is there any difference between a Gen 2.5 and a Gen 3?

yea, the checkering in the finger grooves. The 2.5 finger grooves are smooth, the gen 3 are checkered.
